Antibiotic Resistance: Microfluidic Evolution‐On‐A‐Chip Reveals New Mutations that Cause Antibiotic Resistance (Small 10/2021)

چکیده
Microfluidics reveals ambiguous antibiotic resistance scenarios. A custom microfluidic device reported by Kersten S. Rabe and co-workers (article number 2007166) elucidates for the first time how bacteria turn into either resistant or persistent “superbugs” due to varying exposure. The chip accelerates evolution, leading discovery of previously unknown mutations within hours, paving way a better understanding mechanisms resistance.

Spontaneous mutations that confer antibiotic resistance in Helicobacter pylori.
In this study, we systematically examined in vitro frequencies and spectra of the spontaneous mutations in Helicobacter pylori that confer resistance to clarithromycin (Cla(r)), metronidazole (Mtz(r)), amoxicillin (Amx(r)), ciprofloxacin (Cip(r)), and rifampin (Rif(r)). The mutation rate of Rif(r) or Cip(r) determined in a fluctuation assay is 1 x 10(-8) to 2 x 10(-8) per cell per division. In ...
متن کاملThe fitness costs of antibiotic resistance mutations
Antibiotic resistance is increasing in pathogenic microbial populations and is thus a major threat to public health. The fate of a resistance mutation in pathogen populations is determined in part by its fitness. Mutations that suffer little or no fitness cost are more likely to persist in the absence of antibiotic treatment. In this review, we performed a meta-analysis to investigate the fitne...
متن کاملWhere antibiotic resistance mutations meet quorum-sensing
We do not need to rehearse the grim story of the global rise of antibiotic resistant microbes. But what if it were possible to control the rate with which antibiotic resistance evolves by de novo mutation? It seems that some bacteria may already do exactly that: they modify the rate at which they mutate to antibiotic resistance dependent on their biological environment. In our recent study [Kra...
متن کاملAntibiotic Resistance
Antibiotic resistance is an ongoing evolutionary process that has been the concern of infectious disease specialists since antibiotic use gained popularity. Antibiotic resistance is not a concern of the public as reflected by their desire for antibiotics whenever illness occurs. Patients can and do pressure their physicians for antibiotics ; 12 million antibiotic prescriptions were filled for u...
